Caracterização e resfriamento do sêmen de curimatã, Prochilodus brevis (Steindachnner, 1874)

Resumo

The Prochilodus genus is present in South America, being quite found in Brazilian basins. The aim of this study was to characterize the cryopreserved sperm P. brevis through analysis of physical and chemical parameters of the seminal fluid, the time of sperm motility and the effect of cooling on the morphology of the sperm, and performs tests cooling seminal fluid. Adult organisms were used, weighing on average 295.5 ± 65.9 g and length of 29.0 ± 2.3 cm. The seminal volume collected had a mean of 1.55 ± 0.86 mL. For the average sperm concentration was 1.5 ± 0.3 x 107 spz mL -1, while the average pH was 7.9 ± 0.2. The osmolarity of sperm was 287.9 ± 48.5 mOsm/kg and the rate of sperm motility of sperm average fish was 63 ± 1.6 s. The anomalies were found (A) defect in the anterior region of the sperm (irregularity of the membrane) and the intermediate section with a change in shape with 2.5%; (B) intermediate piece shaped corkscrew 3.0%; (C) intermediate part and tail curled tightly around the head with 1.0% and D) intermediate piece retro-axial position with 6.7% to 13.2% of observed anomalies, occurring statistically significant difference (p>0.05) between the amounts of morphological abnormalities. Calcium ions, chloride and magnesium present mean concentrations of 10.11 ± 0.03 mg / dL, 99.93 ± 0.60 mg / dL and 1.99 ± 0.01 mg/ dL, respectively. Regarding thinners, Ringer's solution showed a significant result after 21 h of cooling (50 ± 23.0%), whereas glucose (0 ± 0%), Sucrose (0 ± 0%) and NaCl (0 ± 0 %) were not efficient. In conclusion, Ringer's solution is indicated to preserve for longer activity of spermatozoa in the sperm of this species.
