Influence of auxins and explants types in the induction of friable callus in Piper hispidinervum C. DC

The objective of this work was to evaluate the influence of auxins and explants types in the callogenesis of Piper hispidinervum C. DC. in cultivation of cells in suspension. As explant source was used leaf and internode segments obtained from plantlets germinated in vitro. The basal medium constituted of the salts and vitamins of MS. As auxins were tested NAA, IBA, IAA and 2,4-D in concentrations of 0; 2.5 and 5 mg L-1. The explants were disposed in 250 mL flasks with 30 mL medium, being the leaf segments inoculated with the abaxial side in contact with the medium. Six explants per treatment were arranged in a completely randomized design with five replications. Cultures were maintained in the dark in a growth room at 25±2 ºC for three weeks, when the callus formation was evaluated. It was verified that the explant type significantly influenced the callogenesis. Foliar explants were more responsive than internodes and for both explants the callus formation was larger when to the culture medium was added NAA.Downloads
