Non-destructive method for extracting DNA from cashew seeds

The accession characterization step is of fundamental importance for both evaluation of a germplasm collection and for a genetic breeding program. Actual plants in the field are needed to permit a characterization step, whether morphological or molecular, to observe overall aspects of the plant or to collect leaves for DNA extraction, a process that demands considerable amounts of time and resources. On the other hand, DNA extraction performed directly on the seed allows the genome to be accessed without the need to plant all seeds, accelerating the choice of genotypes of interest and information on genetic variability. In this sense, the objective of this work was to develop a non-destructive methodology for extracting DNA from cashew seeds. The seeds were mechanically perforated with a mini grinder, and a sample of the endosperm of each seed was taken. For DNA extraction, different protocols and commercial kits were tested, as well as materials to seal the opening made in the seed. In conclusion, it is possible to extract cashew tree DNA from the endosperm, preferably using commercial extraction kits and cement as a sealing material for later planting.
