Molecular polymorphic markers between moco and herbaceous cotton

Abstract

Cotton breeding programs continuously develop varieties to supply producers and textile industry. The aim of this work was to evaluate molecular markers analyzing a population derived from an intercross between the upland cotton variety Codetec 401  and the moco variety CNPA 6M. Seeds from 93 F2 individuals were planted and DNA was extracted, then amplified by 62 microsatelite primer pairs and 33 RAPD primers. Amplified fragments were separated by vertical acrylamide or horizontal agarose gels, stained by silver nitrate or ethidium bromide, respectively, for microsatellite and RAPD. Twenty six microsatellite markers (42%) were polymorphic. A total of 325 RAPD markers were obtained, with a medium number of 9,8 markers per primer. The polymorphic RAPD markers totalized 101 (31%). Intercrosses among moco and upland cotton have a reliable quantity of segregating markers; hence can be used in molecular mapping efforts. Furthermore, for its genetic proximity to upland cotton, moco cotton can be an option to be used on cotton pre-breeding programs, and the markers can be used for genomic selection.
